How does half life of reporter proteins influence transcriptional response? (2)
In this graph, 3 h-pulse with 10 fold increase at mRNA level is assumed, and the response of reporter proteins is calculated.
The graph demonstrates reporters with long half lives can only show poor responses in this case. Especially, detection of down regulation is almost impossible. In contrast, reporters with short half lives can reasonably represent the change of the mRNA level.
